LA 시가지가 한눈에 내려다보이는 멀홀랜드 드라이브 도로를 차 한 대가 미끄러지듯 달려간다. 차의 뒷좌석에는 미모의 여인 리타가 수심에 찬 눈빛으로 앉아 있다. 차가 조용히 멈추고 여자가 의아스러운 표정으로 말한다. "이 길이 아니잖아요?" 차를 몰던 남자가 그녀에게 총구를 들이대는 순간 맞은편에서 달려오던 폭주족 차량이 정면으로 여자의 차를 들이받는다. 여자는 사고 속에서 살아나지만 모든 기억이 사라진 상태다. 그녀가 또다른 여인 베티의 도움으로 기억을 찾아가면서 끔찍한 악몽이 시작되는데...
